Al finalizar el Lavatorio, Cristo tomó nuevamente su manto y se sentó. Después de la cena judía, pidió a los apóstoles que igualmente se sentaran y acomodaran un paño sobre la mesa, que colocaran un plato con un pan de trigo sin levadura y una copa grande con vino. Entre oraciones se dirigió a sus apóstoles diciéndoles que habían sido escogidos para fundar la Iglesia. Entonces consagró el pan dejándolo convertido transubstancialmente (cambiando de una sustancia a otra) en su cuerpo y el vino en su sangre (sacramento de la Eucaristía). Antes de dividir el pan para repartirlo entre sus apóstoles, les anunció su inminente muerte y resurrección; después de eso, les pidió que se dedicaran a predicar la palabra de Dios.
